Four groups of phosphatase activity were found in anaerobic digesting sludge, with pH optima of 4.2, 6.6, 9.4 and 11.4. Increases in acid and especially alkaline phosphatase activity were found in anaerobic digesters overfed to the point of souring and failure. Increased activity was observed up to 10 days before higher molecular weight volatile fatty acids (iso butyric, butyric, iso valeric and valeric) characteristic of digester souring were detected. Phosphatase activity in anaerobic digesters appears to be a rapid biochemical test for predicting the instability or failure of anaerobic sludge digestion.
